Anti-Osteoarthritis Effect of Schisandra chinensis (Turcz.) Baillon Ethanol Extract on H2O2 or LPS-Stimulated Primary Chondrocytes

Abstract
Osteoarthritis is a chronic disease characterized by cartilage and progressive degradation pain. Schisandra chinensis (Turcz.) Baillon is Asian used in traditional medicine to treat heart disease, insomnia, skin irritation, rheumatoid arthritis, cough, and dyspnea. The inhibitory effect of a Schisandra chinensis (Turcz.) Baillon ethanol extract (SB) on osteoarthritis was investigated in primary cultured rat cartilage cells. The pro-inflammatory cytokines (IL-1¥â, IL-6, TNF-¥á), PGE2, and NO were measured by ELISA. The anabolic (aggrecan, collagen type ¥°, ¥±) and catabolic (MMP-3, MMP-13, TIMP-1, and TIMP-3) factors were measured by PCR and western blot analyses and the inflammatory (NF-¥êB pathway, COX-2, iNOS) factors were determined by western blot. SB significantly decreased the follwing: production of pro-inflammatory cytokines, PGE2, and NO; mRNA expression of catabolic factors; and the protein expression of catabolic and inflammatory factors. In addition, SB significantly increased the mRNA expression of anabolic factors. This study suggests that SB is a potential functional food component to control inflammation and osteoarthritis.
